Bilirubin (Jendrassik & Grof) is a reagent kit for the quantitative estimation of total and direct bilirubin in serum or plasma. Bilirubin reacts with diazotised sulphanilic acid to form an azo compound whose colour, read at 546 nm, is proportional to the bilirubin present; caffeine accelerates the reaction for total bilirubin. Manufactured at Reckon’s Vadodara facility under ISO 13485:2016 quality management, and released against reference standards before dispatch.

Procedure at a glance5 steps
- Mix equal volumes of 1-Bilirubin A and B to make working solution 1, and bring reagent 3 to room temperature.
- Total bilirubin: to 50 µl of sample add 100 µl of working solution 1 and 1.0 ml of solution 3; for the sample blank use solution 2 instead of working solution 1.
- Mix, incubate for 5 minutes at room temperature, and read against the sample blank at 546 nm within 10 minutes.
- Direct bilirubin: to 50 µl of sample add 1.0 ml of normal saline and 100 µl of working solution 1 (blank: solution 2), and read after 3 minutes.
- Bilirubin (mg/dl) = (absorbance of sample − absorbance of sample blank) × 26.312.
