The Widal Tube Test (O, H, AH, BH antigen) is a tube agglutination kit for detecting and titrating antibodies to Salmonella in serum, for the diagnosis of enteric fever. Killed suspensions of S. typhi O and H and S. paratyphi A (H) and B (H) are mixed with a serial dilution of the patient's serum and incubated overnight. Manufactured at Reckon’s Vadodara facility under ISO 13485:2016 quality management, and released against reference standards before dispatch.

Procedure at a glance5 steps

For each serum, set 4 rows of 6 tubes (3 × 3/8 inch) in a rack.
Make a master dilution in 5 tubes: 7 ml of normal saline in the first and 3.5 ml in the rest; add 0.5 ml of serum to the first and carry 3.5 ml down the row, for 1:30 to 1:480.
Transfer 0.5 ml from each master tube to the matching tube in every row, and put 0.5 ml of saline in the sixth tube of each row as a control.
Add 0.5 ml of S. typhi O, S. typhi H, S. paratyphi A (H) and B (H) antigen to rows 1 to 4, shake, and incubate at 37 °C overnight (16 to 20 hours).
The titre is the highest dilution with visible agglutination, judged against the saline control.
