The Widal Slide Test (O, H, AH, BH antigen + control) is a slide and tube agglutination kit for detecting antibodies to Salmonella antigens in serum, as a screen for enteric fever. Stained, killed suspensions of S. typhi O and H and S. paratyphi A (H) and B (H) agglutinate with the matching antibodies, and the titre comes from the highest dilution that still agglutinates. Manufactured at Reckon’s Vadodara facility under ISO 13485:2016 quality management, and released against reference standards before dispatch.

Procedure at a glance5 steps

Bring all reagents to room temperature and shake each antigen vial well.
Place 1 drop of undiluted serum in each of 4 circles, and add 1 drop of O, H, AH and BH antigen to circles 1 to 4.
Mix each circle with a separate stick, rock the slide for 1 minute, and look for agglutination.
If any circle agglutinates, titrate: 0.08, 0.04, 0.02, 0.01 and 0.005 ml of serum, each with 1 drop of antigen, give titres of 1:20 to 1:320.
In tubes, dilute the serum 1:20 to 1:1280 in saline, add 1 drop of antigen, and incubate at 37 °C for 16 to 20 hours; the titre is the highest dilution with clear agglutination.
