G-6-PD (Qualitative) is a visual dye-decolourisation test for screening glucose-6-phosphate dehydrogenase deficiency in erythrocytes. G6PD in a red-cell hemolysate generates NADPH, which with PMS decolourises blue DCPIP dye; the time taken to decolourise is inversely proportional to G6PD activity. Manufactured at Reckon’s Vadodara facility under ISO 13485:2016 quality management, and released against reference standards before dispatch.

Procedure at a glance5 steps

Add 0.5 ml of buffer to the co-enzyme substrate vial, shake to dissolve, and use within 10 minutes.
Mix 0.02 ml of whole blood (adjusted for hemoglobin) with 1.0 ml of pre-cooled lysing reagent and keep at 2 to 4 °C for 10 to 15 minutes.
Transfer the hemolysate into the working reagent, shake, overlay with 1.0 ml of inert oil, seal air-tight, and incubate at 37 °C.
Watch for the change from blue to brownish colour from 30 minutes, every 5 minutes.
Normal samples decolourise in 30 to 60 minutes; G6PD-deficient samples take 2 to 24 hours.
